چکیده
Glycolysis emerges as a new therapeutic target for malignancies. The inhibition of glycolytic activator, PFKFB3, repairs tumor endothelial cell function, and normalizing the microenvironment. We aimed to investigate significance PFKFB3 in HCC, effects inhibitor, PFK15, HCC cells cells. Double immunofluorescent staining CD31 tissues revealed that high expression both was significantly correlated with poor prognosis. Multivariate analysis identified an independent prognostic factor. PFK15 suppressed proliferation line vitro. In subcutaneous model cells, growth induced apoptosis. Moreover, treatment vessel normalization, decreasing diameter pericyte attachment improving perfusion. High novel marker HCC. Targeting via might be promising strategy suppressing inducing normalization.

DNA methylation of tumor suppressor genes in hepatocellular carcinoma
The basic unit of chromatin is a nucleosome included an octamer of the four core histones and 147 base pairs of DNA. Posttranslational histones modifications affect chromatin structure resulting in gene expression changes. CpG islands hypermethylation within the gene promoter regions and the deacetylation of histone proteins are the most common epigenetic modifications. The aberrant patterns of...
متن کاملPolyclonal Antibody against Different Extracellular Subdomains of HER2 Induces Tumor Growth Inhibition in vitro
Background: Human epidermal growth factor receptor 2 (HER2) has a crucial role in several malignancies. The extracellular domain of HER2 (HER2-ECD) has been extensively employed as an important target in passive and active immunotherapy. Isolated recombinant prokaryotic HER2-ECD subdomains were previously found to be ineffective in inducing anti-tumor antibody response. Objective: To employ rec...
متن کاملGalectin-1 Inhibitor OTX008 Induces Tumor Vessel Normalization and Tumor Growth Inhibition in Human Head and Neck Squamous Cell Carcinoma Models
Galectin-1 is a hypoxia-regulated protein and a prognostic marker in head and neck squamous cell carcinomas (HNSCC). Here we assessed the ability of non-peptidic galectin-1 inhibitor OTX008 to improve tumor oxygenation levels via tumor vessel normalization as well as tumor growth inhibition in two human HNSCC tumor models, the human laryngeal squamous carcinoma SQ20B and the human epithelial ty...
متن کاملTumor suppressor XAF1 induces apoptosis, inhibits angiogenesis and inhibits tumor growth in hepatocellular carcinoma
X-linked inhibitor of apoptosis (XIAP)-associated factor 1 (XAF1), a XIAP-binding protein, is a tumor suppressor gene. XAF1 was silent or expressed lowly in most human malignant tumors. However, the role of XAF1 in hepatocellular carcinoma (HCC) remains unknown. In this study, we investigated the effect of XAF1 on tumor growth and angiogenesis in hepatocellular cancer cells. Our results showed ...
متن کاملSmall-molecule inhibition of 6-phosphofructo-2-kinase activity suppresses glycolytic flux and tumor growth.
6-phosphofructo-1-kinase, a rate-limiting enzyme of glycolysis, is activated in neoplastic cells by fructose-2,6-bisphosphate (Fru-2,6-BP), a product of four 6-phosphofructo-2-kinase/fructose-2,6-bisphosphatase isozymes (PFKFB1-4). The inducible PFKFB3 isozyme is constitutively expressed by neoplastic cells and required for the high glycolytic rate and anchorage-independent growth of ras-transf...
